It would appear that when LR introduced the HSE Luxury c.2012 they used it as a cost saving exercise too, as the HSE had a “standard“ 11-speaker system. Unless you have the Premium Surround Sound system (standard on HSE Luxury and run-out Landmark models with a 17-speaker system including sub-woofer, initially by Harmon-Kardon, and latterly by Meridien) you won’t have a sub-woofer as standard.
Our previous MY2010 HSE with the Harmon-Kardon set-up had the full system, and when I was looking to change I looked at a HSE, but as music (classical) is very important to me, I fund the standard system very weak when compared to the MY2010. One of the reasons I went for the Landmark even though things like the RSE have never been used.
Obviously, the HSE could have been upgraded by ticking the option for the Premium Sound System.

This is also wrong.
All colour screen D4s have a subwoofer. Now not all subwoofers / amps are equal and as you say maybe the premium would have come with beefier models but they all have them (if you have the colour screen)

In the brochures, it specifically states that the premium system has a sub-woofer which isn’t mentioned in the standard system. Perhaps it’s a better model as you suggest? In my experience, the standard set-up might as well have not bothered.
One problem of the “better”(?) sub-woofer is that it shows the less than perfect build quality at times as low organ notes get everything rattling!
I’ve just had a look at the specs again, and you’re quite right. The extra speakers are in the doors and rear pillars on the higher spec system.

I have D4 2.7 2011 GS with the same problem. No sub but I see around black mark on the rear tailgate.Defender 90 TDI300 1996

but you have a GS and unless you have the upgraded amp under your drivers seat you won't have a sub (even with the amp not 100% sure on a GS) unless of course you have the colour screen and then you should have one
big job updating a GS to have an amp and colour screen:

Just had a look in my lower tailgate on my 2013 MY D4, nothing there, just two 2 way connectors. (Purple & Yellow) ( Brown and grey) I believe it was removed by the original owner given a former role of this vehicle; there is evidence someone has been in here before. What do I need to make give me a sub woofer; a speaker or an amplifier and a speaker?

Yes.
I have a sealed 10" Sub with its own amp fed from the output of the factory LR amp that would normally go to the tailgate speaker (which is shocking as you have all discovered!).2015 SDV6 Commercial

I detected some electrical activity on the subwoofer cables when I switched on the unit. I'm wondering the LR unit looks for a subwoofer before powering up the amp. I haven't looked at the amp in the driver's side panel yet but the circuit diagram indicates a sub woofer amp within the KH amplifier. I have a used LR4 subwoofer on its way. It didn't cost a lot so I'll see what happens.

the sub in your vehicle must be blown. it definitely has one. check in the lower boot door
