Hi, I currently run 18" wheels with 255/60 tyres. If I run Cooper ST 245/70s with the above 255/60 as a spare, will there be transmission problems if I need to use the spare when the Coopers are on?

The 245/70 is slightly oversize (standard tyre for the 17" rim is 235/70) so I don't know if you should risk it, probably okay for short journeys though.
If you want another 17" rim to put another Cooper on so you've got a correct sized spare I've got one going spare that you could have for a nominal sum
Speaking of 17" rims and Coopers, I've got a set of 5 with Cooper STTs on that will be up for sale very soon if anyone's interested The End
